About the Job

We are seeking an experienced researcher to participate in studies in transplant immunology and organ cryopreservation. We focus on two areas of research 1) tolerance in organ transplantation and 2) organ cryopreservation. For our tolerance studies, we use regulatory T cells (Treg) to prevent rejection of organs. We have developed several promising approaches and are beginning translational studies in humans. The second area of study is using novel approaches for the long-term preservation of organs prior for transplant. In collaboration with faculty in biomedical and mechanical engineering we have developed a new methodology for banking organs. Our lab focuses on the biologic assessment and in vivo function of these organs after recovery from cryopreserved state. This position will participate in both areas of research. Research involves study of human tissues and the use of translational animal transplant models.

Specific responsibilities include but are not limited to:
Lab Management (10%):
Ensuring the laboratory is properly stocked. Routine lab maintenance.

Coordinate the use and maintain the upkeep of expensive specialized laboratory equipment; including PCR machines, electrophoresis gel imager, centrifuges, sterile culture hoods, cold storage systems, and sterile incubators.

Study design and execution (80%):
Participate, under supervision, in the planning of experiments designed to address biologic questions in the two areas of study. (Essential skills: experience in design and planning of experiment using existing protocols, completing experiment, recording data, and summarizing conclusions).

Isolate and expand specific T cell types (Treg). This involves routine cell isolation (magnetic bead separation), extensive cell culture, performing T cell suppression assays (MLR assays), and FACS phenotyping of cells. (Essential skills: cell culture, flow cytometry. Desired skills: T cell subset isolation, T cell assays, Intracellular FACS).

Routine molecular biology for producing modified plasmid DNA, production of lentivirus, and transduction of T cells. (Essential skills: PCR, molecular cloning, gel electrophoresis, plasmid purification. Desired skills: Lentivirus production, RT-PCR, qPCR, cell transfection).

Viability and functional assessment of tissues after cryopreservation. (Essential skills: viability staining. Desired skills: fluorescent microscopy, tissue sectioning, ELISA, Seahorse assay).

Assist in small animal transplant studies. Assist in performing transplants and recovery of tissues for study. (Essential skills: preparation of solutions with defined concentrations and pH. Desired skills: experience with small animal colony management, injection, tissue recovery, islet purification)

Data analysis and Presentation (10%):
Analysis and presentation of data to supervisors, immediate work unit and in collaboration with other work groups in the department of surgery and joint lab group.

Reading relevant publications on transplant tolerance and organ cryopreservation and participating in discussion of project development.
